چکیده

Hematopoietic progenitor cells migrate in vitro and in vivo towards a gradient of the chemotactic factor stromal cell-derived factor-1 (SDF-1) produced by stromal cells. This is the first chemoattractant reported for human CD34 1 progenitor cells. Concentrations of SDF-1 that elicit chemotaxis also induce a transient elevation of cytoplasmic calcium in CD34 1 cells. SDF-1-induced chemotaxis is inhibited by pertussis toxin, suggesting that its signaling in CD34 1 cells is mediated by seven transmembrane receptors coupled to G i proteins. CD34 1 cells migrating to SDF-1 include cells with a more primitive (CD34 1 /CD38 2 or CD34 1 /DR 2 ) phenotype as well as CD34 1 cells phenotypically committed to the erythroid, lymphoid and myeloid lineages, including functional BFU-E, CFU-GM, and CFU-MIX progenitors. Background and Objective: Mobilization of Hematopoietic Stem Cells (HSCs) for transplantation and the importance of -adrenergic signals in induction of this process have been well investigated. However, little is known about the role of -adrenergic signals in mobilization of HSCs and factors influenced by these signals. The Chemokine Stromal Derived Factor -1 (SDF-1) which is expressed by hum...

Advances in Hematopoietic Stem Cell Mobilization and Peripheral Blood Stem Cell Transplantation

Hematopoietic stem/progenitor cells (HSPCs) which give rise to different blood cell types are present within the bone marrow microenvironment, especially in flat bones such as skull, vertebrae, pelvis and chest. Interacting factors such as stromal derived factor-1/CXCR4, very late antigen-4/vascular cell adhesion molecule-1, Lymphocyte function-associated antigen-1/ intercellular adhesion molec...

Sulfated polysaccharides increase plasma levels of SDF-1 in monkeys and mice: involvement in mobilization of stem/progenitor cells.

It was previously reported that treatment with the sulfated polysaccharide fucoidan or the structurally similar dextran sulfate increased circulating mature white blood cells and hematopoietic progenitor/stem cells (HPCs) in mice and nonhuman primates; however, the mechanism mediating these effects was unclear. It is reported here that plasma concentrations of the highly potent chemoattractant ...

In vitro behavior of hematopoietic progenitor cells under the influence of chemoattractants: stromal cell-derived factor-1, steel factor, and the bone marrow environment.

How multiple chemoattractants cooperate in directing the migration of hematopoietic progenitor cells (HPC) for homing and peripheral blood mobilization has not yet been established. We report here the behavior of HPC under the influence of two different chemoattractants, stromal cell-derived factor (SDF)-1 and steel factor (SLF), and the chemotactic nature of the bone marrow (BM) environment us...
